IIS restriction enzymes have already found their way into many innovative cloning and DNA assembly protocols. However, the assembly of up to 9 fragments into a defined order in a single reaction is a nice advance. The new protocol looks promising not only for the generation of libraries but could also prove useful for synthetic biologists that try to assemble constructs from many modules.

This cloning strategy could be listed among the DNA assembly standards being evaluated by the BioBrick Foundation http://bbf.openwetware.or....
