The authors present a very unique computational method that models HS domain structure based on the relatively small amount of information provided by disaccharide analysis data. This approach will be very useful for proteoglycanologists studying structure-function relationships, especially since it is currently a very laborious task to determine the organization of the various domains of HS that are responsible for the functions of this essential macromolecule. It would be of significant interest to apply the methods outlined in this manuscript to the modeling HS from cells that have been genetically manipulated with respect to their expression of enzymes that modify HS during biosynthesis (e.g. 2O-sulfotransferase) or subsequent to biosynthesis (e.g. the Sulfs).
