Reviewer 3's Review (Steven de Belle)

“This is a manuscript of significance that reports the effects of genetic, physiological and pharmacological manipulations on thermotolerance in Drosophila melanogaster and Locusta migratoria. Strengths of the work include (1) use of multiple assays (behavior, electrophysiology), demonstrating robust effects of experimental manipulations, (2) use of multiple manipulations (heat pre-exposure, biochemical intervention) to dissect hyperthermia response mechanisms, (3) parallel findings using similar manipulations in a genetic model (fly) and physiological model (locust), (4) the important conclusion that cellular events involving PKG and PP2A may be intimately connected with heat tolerance mechanisms, and (5) the ecological relevance of the findings.”
