“This is an important and interesting paper. Previously, the authors have reported that an artificial zinc finger 'Jazz' binds to the utrophin promoter and activates its transcription. Here they further validate the same system in vivo using the transgenic approach. They established the zinc finger artificial transcription factors (ZF ATFs) "VP16-Jazz" transgenic mice under the control of the myosin light chain promoter. The expression of "VP16-Jazz" in transgenic mouse up-regulates its target gene "utrophin" at the chromosomal site, which was tested by chromatin immunoprecipitation experiments. This is an excellent example demonstrating that the artificial zinc finger transcription factor works in mice and lacks toxicity.”
