The use of MFI as a metric occurs strange to me, since
the figures clearly demonstrate distinct subpopulations (i.e. STAT3+/5+) and they also display the percentages on each plot. Wouldn't it be better to use the percentages of well characterized cell populations - after all, this is the power of cytometry in single cell analysis?

Comment 2.
The blast 1 subpopulation in figure 6 seems to be comprised by immature precursors (myelocytes and so on) rather than "real" blasts. At least morphologically CD45brightCD33+ cells are usually not blasts, although they probably belong to the malignant clone (depending on the subtype of AML and ...the leukemogenesis model that one supports).
