Review of the original submission:

Zen and colleagues present the results of a series of studies that were designed to investigate the roles of e-selectin and CD44v4 in transendothelial migration of a battery of human breast cancer cell lines. The authors report that migration of breast cancer cells across HUVEC monolayers is dependent on e-selectin, and that CD44v4 serves as an important ligand for e-selectin on the breast cancer cell.

The studies were carefully designed, and their sequential conduct and reporting provides a convincing body of work to support their conclusions.

Minor Comments:

1. The HUVEC model provides a highly simplified experimental model for the study of tumor cell invasion. Addition of a brief discussion of the strengths and weaknesses of this model would be useful to readers whose work lies outside of the area of invasion and metastasis.

2. Although all of the cell lines used are well-studied, it would be useful for the authors to include a brief comment identifying the rationale for the selection of each cell line for use in the study.

3. Figure 1 is redundant of the text and should be considered for deletion. Alternatively, a minor modification of the text (so that specific data are presented only in the Figure) should be considered.
